Which version of dreamup do you use? And which db are you trying to flash?

as baluba says which version of dreamup are you using and which version of dreambox do have ,are you using a usb to serial adaptor with the null modem cable .if your using an adaptor have you installed a driver for it (a driver is a bit of software that may come on a disk with the adaptor or you download it and simply put it helps the computer to see the box so it can be flashed) if your pc runs on vista it can be problematic with drivers. if your box is a dm500 i would say use an older version of dreamup (have a look in the links in my signature area) in my experience if the version of dreamup was the issue it would give an error message like flash failed aborted by user ,if you click connect and nothing happens its usually either the wrong com port and most often the driver as i mentioned above .
i love it when a dreambox comes together.
