You should only delete programs from within the "add/delete programs" reached through the control panel.

You can also do a disk cleanup, which you can get to from the start menu and system tools. which will allow you to delete cookies, temp files and empty your trash bin etc

            Also do some spyware/virus checking.

There are some nasty things arround.. still.. there can be many causes for the slowdown including but not only fragmentation, heavy AV software, loads of crap running on the background of your OS.. anyway lots of things.

If you do that important things that should start (like your antivirus) will not run, look to see what crap is starting with windows and disable it

download superantispyware ( its free) and do a complete system scan
ccleaner as Dasteph suggests is one I use as well for clearing all the crap out of your system, it also incorporates a registry cleaner

another thing to do is go my computer, click c drive, click search and select the photos and video selection, after the search is compleet look at all your pictures you want to keep along with all your videos and copy them to a disc, you can do the same wyth your music, after that format your drive and do a new instalation,it should now be like new,,mind and make sure you have all the drivers that you need to get it running again, good luck

Advanced systemcare is also a good scanner, and it scan for spyware, registry, privacy sweep and useless files.

You could also try removing unnecessary windows components via 'add & remove programs' in the control panel.
You will be surprised how much stuff is installed that you don't actually use.
